چکیده

The distribution of j3-adrenergic receptors in sections of the human right and left coronary arteries and of the anterior intraventricular branch was studied by the use of combined in vitro radioreceptor binding and autoradiographic techniques. [125I]Cyanopindolol was used as a ligand for ,B-adrenergic receptors. Binding of the radioligand to sections of the three coronary arteries under study was saturable, stereoselective, reversible, and displaceable by antagonists and agonists with the rank order of potency expected for j3-adrenergic receptors. Analysis of binding isotherms indicated maximum binding capacities of 41.5 fmol/mg protein for the right coronary artery, 35.4 fmol/mg protein for the left coronary artery, and 25.7 fmol/mg protein for the anterior interventricular branch. Dissociation constants were -35 pM in the arteries examined. The relative amounts of j81and 2-receptor subtypes were as follows: 72% p1-receptors and 28% 32-receptors in the right coronary artery; 65% j31-receptors and 35% p2-receptors in the left coronary artery; 40% p3-receptors and 60% P2-receptors in the anterior interventricular branch. The results of autoradiographic analysis revealed a predominance of f31-receptors in the medial layer. p2-Receptors were localized primarily in the adventitia, in the adventitia-media border, and in the intimal layer. These results should lead to a better understanding of the mechanisms involved in the control of coronary circulation in humans. (Circulation Research 1991;68:1591-1599)

Quantitative Autoradiographic Delineation of the Distribution of /3-Adrenergic Receptors in Canine and Feline Left Ventricular Myocardium

The distribution of adrenergic receptors in specific components of the heart such as vessels and myocytes cannot be determined easily with assays of membranes prepared from homogenates of whole tissue. Accordingly, we characterized the binding of the potent nonsubtype selective antagonist [iodo]cyanopindolol to /3-receptors in unfixed transmural slices of feline and canine left ventricle. Speci...

Subtypes of /3-Adrenergic Receptors in Bovine Coronary Arteries

Whether large coronary artery dilation induced by /3-adrenergic stimulation is mediated by /3ior /32adrenergic receptors remains controversial. This problem is particularly difficult to address in vivo due to the concomitant increase in coronary blood flow with /3-adrenergic stimulation, which by itself can dilate large coronary arteries. To reconcile this problem, 5 calves were instrumented wi...

Autoradiographic mapping of calcitonin gene-related peptide receptors in human and guinea pig hearts.

Calcitonin gene-related peptide (CGRP) is a 37-amino acid peptide that is a potent coronary vasodilator. Although CGRP is found in high concentrations around coronary arteries, its precise function in the control of coronary vasomotor tone remains unclear. We studied the distribution of specific receptors for CGRP in guinea pig and human hearts and found that the highest concentration of specif...
